Test Principle of the IDK® Beta-Klotho ELISA

Assay standards and pre-diluted samples assayed for β-klotho are added into the wells of a microplate coated with an anti-β-klotho antibody. During the first incubation step, klotho is bound to the plate by the immobilized primary antibody. Then, a biotin-labeled anti-β-klotho antibody is added.

In the next step, the streptavidin-peroxidase-labeled conjugate is added into each microtiter well, and the following complex is formed at the wall of the plate: primary antibody – β-klotho – biotinylated antibody – streptavidin-peroxidase-conjugate.

Tetramethylbenzidine (TMB) is used as a peroxidase substrate. Finally, an acidic stop solution is added to terminate the reaction. The color changes from blue to yellow. The intensity of the yellow color is directly proportional to the concentration of klotho.

A dose-response curve of the absorbance unit (optical density, OD at 450 nm) vs. concentration is generated using the values obtained from the standard. The presence of beta-klotho in the samples is determined directly from this curve.
