Test Principle of the IDK® anti-htTG IgA ELISA

The IDK® anti-htTG IgA ELISA is a sandwich assay for the quantitative determination of anti-tissue transglutaminase antibodies (IgA anti-htTG).

The wells of the microtiter plate are coated with the antigen. In the first incubation step, the anti-htTG IgA antibodies are bound to the coated antigen. To remove all unbound substances, a washing step is carried out.

In a further incubation step, a peroxidase-labeled antibody is added. After another washing step to remove all unbound substances, the solid phase is incubated with the peroxidase substrate, tetramethylbenzidine (TMB). Finally, an acidic stop solution is added to terminate the enzymatic reaction, whereby the color changes from blue to yellow. The intensity of the yellow color is directly proportional to the IgA anti-tissue-transglutaminase antibody concentration.

A dose-response curve of the absorbance unit (optical density, OD at 450 nm) vs. concentration is generated using the values obtained from the standards. The presence of IgA anti-tissue transglutaminase antibodies in the samples are determined directly from this curve.
