The first step in determining coenzyme Q10 includes sample preparation. Therefore, the internal standard is added to the calibrator, controls, and samples. Higher molecular substances are removed by precipitation and centrifugation.
The coenzyme Q10 in the supernatant is extracted by an organic solvent, evaporating afterward. The residue is resuspended in ethanol p.a. Afterward, 100 μl of the resuspended solution is injected into the HPLC system.
The separation via HPLC follows an isocratic method at 30 °C using a reversed-phase column. One run lasts 15 minutes. The chromatograms are recorded by a UV detector. The quantification is performed with the delivered calibrator; the concentration is calculated via integration of the peak areas/heights by the internal standard method.
