Description

The IDK® alpha-1-Antitrypsin ELISA is an enzyme immunoassay intended for the quantitative determination of alpha-1-antitrypsin in stool.

For research use only. Not for use in diagnostic procedures.

For Laboratory Professional Use Only.

The IDK® alpha-1-Antitrypsin ELISA is compatible with our IDK Extract® Stool Sample Preparation System.

If your lab is testing at high volumes, the IDK® α1-Antitrypsin ELISA is also available in a customized bulk pack (20 plates).

This is the RUO version of Immundiagnostik part number K 6750.

Test Principle of the IDK® alpha-1-Antitrypsin ELISA

The IDK® alpha-1-Antitrypsin ELISA is designed for the quantitative determination of α1-antitrypsin in stool. The assay utilizes the sandwich technique with two selected antibodies that bind to human α1-antitrypsin. Standards, controls, and prediluted samples assayed for human α1-antitrypsin are added into the wells of a microplate coated with a high affine anti-human α1-antitrypsin antibody.

During the first incubation step, α1-antitrypsin is bound by the immobilized antibody. Then a peroxidase-conjugated polyclonal anti-human α1-antitrypsin antibody is added into each microtiter well, and a sandwich of capture antibody – human α1-antitrypsin – peroxidase-conjugate is formed. Tetramethylbenzidine is used as peroxidase substrate. Finally, an acidic stop solution is added to terminate the reaction. The color changes from blue to yellow. The intensity of the yellow color is directly proportional to the concentration of α1-antitrypsin.

A dose-response curve of the absorbance unit (optical density, OD at 450 nm) vs. concentration is generated using the values obtained from the standard. α1-antitrypsin present in the samples is determined directly from this curve.
